NHS Launches Global PrevenTRON Trial of Trontinemab to Prevent Alzheimer's Dementia Before Symptoms
The global Phase 3 study, known as PrevenTRON, is evaluating the experimental drug trontinemab from Roche. This trial involves 1,600 participants aged between 55 and 80 across 18 countries, all of whom have elevated levels of p-tau217, a biomarker associated with cognitive decline. Prior studies indicate that p-tau217 testing can accurately diagnose Alzheimer's disease in 95% of cases, with 78% of diagnosed individuals likely to experience cognitive impairment within ten years. Trontinemab is designed to eliminate amyloid plaques that harm brain cells. In the Phase 2 trial, it successfully removed plaques in 90% of 100 early-stage Alzheimer's patients in just 28 weeks. Phase 3 results are anticipated by 2028, and the trial is led by Professor Ramin Nilforooshan, focusing on pre-symptomatic intervention. Further information can be found on the Alzheimer's Society website.
Key facts
- The PrevenTRON study is a global Phase 3 trial testing trontinemab in people without Alzheimer's symptoms.
- Trontinemab is an experimental drug developed by Swiss pharmaceutical company Roche that targets amyloid plaques.
- The trial involves around 1,600 participants aged 55 to 80 from 18 countries with elevated p-tau217 in their blood.
- Blood tests measuring p-tau217 can identify Alzheimer's disease with up to 95% accuracy.
- About 78% of people with the p-tau217 biomarker could develop cognitive impairment within 10 years.
- In Phase 2, trontinemab cleared amyloid plaques in nine out of 10 people with early-stage Alzheimer's within 28 weeks.
- Results from the current Phase 3 trial are expected no earlier than 2028.
- Professor Ramin Nilforooshan is consultant psychiatrist and chief investigator for the PrevenTRON trial.
- In 2022, lecanemab became the first drug shown to slow Alzheimer's progression, and donanemab produced similarly promising results.
